What is horse serum used for?
Gibco horse serum is donor-herd collected in New Zealand and can be used as an alternative to fetal bovine serum.Horse serum has been cited for use in several specific cell culture applications:Support of in vitro antibody response to SRBC in place of FBS [1]Production of C2 heterokaryons [2]Isolation and culture of primary neural cells [3]Promotion of myogenic differentiation [4]Supplement in mycoplasma broths and agars [5]In addition to horse serum as a lower cost alternative to FBS, Thermo Fisher Scientific also offers normal horse serum that can be used as a blocker solution in immunoassays.Gibco horse serum is available in both heat-inactivated and non-heat-inactivated formats.
Horse serum vs FBS
Horse serum can be used as a viable FBS alternative in some cell culture applications. One key horse serum benefit compared to FBS is its lower cost. However, horse serum composition does differ from FBS composition, with horse serum tending to exhibit lower growth factor and higher immunoglobulin levels than FBS [6]. Therefore, due to differences in FBS and horse serum, your specific experiment and needs should be carefully considered when deciding whether to use horse serum.
Horse serum collection, manufacturing, and testing
Collected from donor herds in New Zealand, each lot of horse serum is tested for its ability to support the growth of Sp2/O-Ag14 (murine myeloma) cells in control medium containing the test lot of serum. Results are compared to those obtained using control growth medium with a previously characterized reference serum. In addition, Gibco horse serum is EIA (equine infectious anemia) tested.

血清如何保存, 可以放多久?
贮藏温度≤-15℃,可长时间保存; 2~8℃存放时,请勿超过一个月。
血清溶解以后,出现沉淀怎么办?
若您欲去除这些絮状沉淀物,可以将血清分装至无菌离心管内,以400-600g离心5min,上清液即可加入培养基内一起培养。 我们不建议您以过滤的方法去除这些絮状沉淀物, 一方面它可能会阻塞您的过滤膜;另一方面,过滤血清这种行为可能会导致血清中部分营养成分的流失。

细胞培养中“低调的高手”——马血清 - 知乎切换模式写文章登录/注册细胞培养中“低调的高手”——马血清灵儿细胞培养中的马血清说起细胞培养用的血清,大多数人都会想到胎牛血清、新生牛血清,对于马血清却知之甚少。今天就给大家介绍一下血清中“低调的高手”——马血清。一、马血清作用细胞培养级的马血清,经过多次过滤,去除了许多大分子蛋白,但保留了血清中促细胞生长的活性因子,表现出与胎牛血清和牛血清的类似的特性,因而能用于细胞培养。马血清可以刺激成纤维细胞分化,以及细胞外基质 (ECM) 、细胞因子的合成。另外培养神经细胞和心肌细胞、诱导生成树突状细胞等实验均可用马血清替代胎牛血清。因此,马血清通常适用于那些研究有丝分裂后,细胞关于生长因子和激素等问题的实验。或许,我们可以这样说,从现有研究来看,马血清更适合用来研究特定种类细胞的分化。实际实验应用方面:马血清已被用作脑皮质神经元-胶质细胞培养的一种必添加物;大鼠的 PC12 细胞,在培养过程中,也需要在基础培养基中添加马血清作为补充剂。1. 马血清可用于细胞培养和干细胞的诱导分化,例如:(1)RPMI-1640培养液加马血清培养脐带血单个核细胞。(参考文献:李春莉, 王建伟, 姜蓉, et al. 人参总皂苷诱导红系血细胞增殖的信号转导[J]. 中国组织工程研究, 2009(23):4568-4572.)(2)原代培养神经细胞。有人用含10%马血清的DMEM,分离培养BALB/c小鼠原代脑神经细胞。发现和添加2%神经细胞生长剂B27的Neurobasal培养基相比,含10%马血清的DMEM培养基更节省成本,方便廉价。(参考文献:李丹, 吕耀中, 王微,等. 两种培养基分离培养BALB/c小鼠原代神经细胞方法的比较[C]// 中国北方实验动物科技年会. 辽宁省实验动物学会, 2015.)(3)诱导脂肪干细胞向成肌细胞分化(文献来源:刘杰,傅强.马血清在脂肪干细胞成肌诱导中的作用[J].中国组织工程研究, 15(14))(4)培养骨髓基质细胞。与胎牛血清相比,马血清对小鼠骨髓基质细胞的促增殖作用更好。(文献来源:王改琴. 当归多糖对小鼠骨髓基质细胞黏附分子表达及其黏附功能的影响[D]. 学位论文.重庆医科大学, 2008.)(5)添加20%马血清的L-DMEM培养液可诱导人骨髓间充质干细胞分化为脂肪细胞。(文献来源:赵文静,陈亚洁,刘薇.人骨髓间充质干细胞分离培养及生物学特性研究.中国现代医学杂志.2006.16(18):2744-2747)2. 马血清添加到微生物培养基中,可用于培养微生物,如:(1)马血清添加到培养基中用于支原体的培养。在《兽药典》中,含马血清的支原体培养基被用于兽用疫苗的支原体检查。用KM2培养基(含马血清)还可培养猪肺炎支原体。(文献来源:胡茂志 焦新安,等.猪肺炎霉形体单克隆抗体的制备与鉴定.《中国兽医科技》 2004年12期)(2)马血清加入到固体和液体培养基中,可用于培养幽门螺杆菌。并且,和羊血清和牛血清比较,添加马血清的培养基更易于分装和保存。(文献来源:刘琳娜, 丁士刚, 张静,等. 几种培养幽门螺杆菌方法的比较[J]. 胃肠病学, 2012, 017(004):240-241.)3. 马血清及激素可共同诱导建立家兔股骨头缺血坏死的模型。(文献来源:洪嵩, 张天宏. 非创伤性股骨头坏死模型的血液流变学及脂质代谢[C]// 2009第一届贵州骨科论坛.)二、注意事项与胎牛血清一样,马血清在使用过程中也应注意几个原则:1、避免反复冻融;2、三步法解冻,及时分装;3、配制好的完全培养基在4℃储存不要超过7天。还有一个大家比较关心的问题,马血清是否需要灭活?我们都知道,胎牛血清由于特殊的采血时间(5-8月胎龄),牛胚胎体内还没有形成完整的补体系统,因而不需要采用热灭活的手段对补体进行灭活。而马血清,在没有特殊说明的情况下,一般采自成年马匹。作为脊椎动物,成年马也拥有完整的补体系统。因而,如果所进行的实验中,涉及补体相关内容,建议大家在使用前对马血清进行热灭活,也可以直接购买商品化的灭活马血清。一些比较成功的实验案例中,所用到的马血清,也是经过热灭活的。相关实验请参考:https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5111218/Ausbian马血清为细胞培养级。它来自健康马血液,为封闭系统内无菌采血过滤而成,为淡黄色澄清液体,超低内毒素,无溶血,无异物,无细菌、真菌、支原体、噬菌体等。产品描述1、每个批次均具有完整的检测报告,其中含有下列检测:1)理化检查包括外观、pH值、渗透压、血红蛋白、总蛋白、内毒素、蛋白颗粒度等2)无菌检查含细菌真菌检查、支原体检查、噬菌体检查,确保不含这些外源因子。3)病毒检查牛病毒性腹泻病毒(BVDV)为阴性。2、极低内毒素。3、提供试用,养细胞更安心。发布于 2022-05-15 21:58细胞死亡细胞细胞学赞同 3添加评论分享喜欢收藏申请

马血清给特别滴细胞特别滴“爱” - 知乎切换模式写文章登录/注册马血清给特别滴细胞特别滴“爱”absin爱必信-absin,中国人自己的生命科学百宝箱Σ众所周知,胎牛血清是细胞培养界的“香饽饽”,备受胞胞们的“宠爱”,其实还有一个“香煎饼”,大家可知道?对一些“挑食”的细胞,马血清可谓是散发着一股“煎饼的清香”。图1 马血清优级(abs989)一、马血清的第一大功能是“滋养”一些特别的细胞,如免疫细胞类、成纤维细胞类、上皮细胞类、神经细胞类等。需要添加马血清的常见细胞类型如下:1)NK(人外周血自然杀伤细胞)和NK-92(人恶性非霍奇金淋巴瘤患者的自然杀伤细胞)营养体系:74% α-MEM基础培养基(abs9506)+ 0.2mM Inositol(abs816057)+ 0.1mM β-mercaptoethanol(abs9592)+ 0.02mM Folic Acid(abs812845)+ 100-200U/mL recombinant IL-2(abs04045)+ 12.5 %马血清优级(abs989)+ 12.5% 胎牛血清优级(abs972)+ 1% 双抗(abs9244)2)EL-4(小鼠胸腺淋巴瘤细胞)营养体系:89% DMEM高糖基础培养基(abs9483)+ 10% 马血清优级(abs989)+ 1% 双抗(abs9244)3)L929(小鼠成纤维细胞)和CRFK(猫肾细胞)营养体系:89% MEM基础培养基(abs9503)+ 10% 马血清优级(abs989)+ 1% 双抗(abs9244)4)MCF 10A(人正常乳腺上皮细胞)营养体系:94% DMEM/F12基础培养基(abs9561)+ 5% 马血清(abs989)+ 1% 双抗(abs9244)+ 20ng/ml EGF(abs04176)+ 0.5ug/ml 氢化可的松Hydrocortisone(abs814512)+ 100ng/ml 霍乱霉素choleratoxin+10ug/ml 胰岛素insulin(abs42225219)5)PC-12(Undifferentiated)(大鼠肾上腺嗜铬细胞瘤细胞(未分化))营养体系:79% RPMI1640基础培养基(abs9484)+ 15% 马血清(abs989)+ 5% 胎牛血清优级(abs972)+ 1% 双抗(abs9244)二、马血清第二大功能是诱导分化作用,有文献报道【1】马血清在 5-氮杂胞苷诱导脂肪干细胞成肌细胞实验中发挥重要作用。该研究如下:将第1代长满100 mm培养皿的脂肪干细胞传代至3个6孔板,实验组培养基中加入体积分数为 5% 马血清 +10 μmol/L 5-氮杂胞苷;对照组培养基中单纯添加 10 μmol/L 的 5-氮杂胞苷;空白组为单纯低糖 DMEM 培养基,其余培养、传代及鉴定所需的条件相同。每日光镜观察记录形态,在培养的第 7、14、28、35 天行免疫荧光和流式细胞仪检测肌蛋白表达差异。该文献的结果与结论是免疫荧光检测成肌特异性胞质蛋白表达以及流式细胞检测相应蛋白表达率提示,加入马血清后的实验组脂肪干细胞的成肌速度和诱导成肌所需时间明显比单纯加入 5-氮杂胞苷的对照组更有优势。实验初步表明马血清在促进脂肪干细胞诱导成肌方面起着缩短诱导成肌所需时间的作用。同时,马血清在诱导 C2C12(小鼠成肌细胞)分化为多核肌管方面也发挥重大作用。C2C12 细胞是由 D·Yaffe 和 O·Saxel 建立的小鼠成肌细胞株的亚克隆;该细胞分化很快,形成可伸缩的肌管并生成特征性的肌蛋白。将状态良好的骨骼肌细胞以 0.9×105 接种在96孔板上(每孔100ul),置于培养箱中培养 24h 后,待细胞融合到 70-80% 后,更换成的含有 2% 马血清的完全培养基中,进行诱导分化。(如图2所示)图3 C2C12(小鼠成肌细胞)向多核肌管方向分化另外,也有文献报道【2】传代培养人骨髓间充质干细胞(P3)在 20% 马血清的 L-DMEM 培养液中可分化为脂肪细胞。此外, 10% 马血清可代替 FBS 用于支持绵羊红细胞的体外抗体应答; 血清 IgG 制备,白蛋白的提取及生物活性研究;免疫组化(IHC)、酶联免疫(ELISA)等免疫实验的封闭剂。参考文献:【1】刘杰,傅强.马血清在脂肪干细胞成肌诱导中的作用[J].中国组织工程研究, 15(14)【2】赵文静,陈亚洁,刘薇.人骨髓间充质干细胞分离培养及生物学特性研究.中国现代医学杂志.2006.16(18):2744-2747本期讲解就到这里了,如有细胞培养问题,欢迎微信公众号后台留言~注:图源网络和客户,仅供参考学习用表 细胞培养类产品推荐(点击查看更多产品信息)货号品名规格abs981胎牛血清(特级)500mlabs972胎牛血清(优级)500mlabs974胎牛血清(标准级)500mlabs9483高糖DMEM培养基500mlabs9484RPMI-1640培养基500mlabs9505MEM培养基500ml发布于 2023-05-05 10:59・IP 属地上海细胞培养NK小鼠细胞赞同 1添加评论分享喜欢收藏申请
